  Plant metabolism is a complex set of processes that produce a wide diversity of foods, woods, and medicines. With the genome se-quences of Arabidopsis and rice in hands, postgenomics studies integrating all ``omics'' sciences can depict precise pictures of a whole-cellular process. Here, we present, to our knowledge, the first report of investigation for gene-to-metabolite networks reg-ulating sulfur and nitrogen nutrition and secondary metabolism in Arabidopsis, with integration of metabolomics and transcriptom-ics. transcriptome and metabolome data suggested the presence of general responses to sulfur and nitrogen deficiencies. In addition, specific responses to either sulfur or nitrogen deficiency were observed in several metabolic pathways: in particular, the genes and metabolites involved in glucosinolate metabolism were shown to be coordinately modulated.   While membrane transporters mediating ammonium uptake across the plasma membrane have been well described at the molecular level, little is known about compartmentation and cellular export of ammonium. AtTIP2;1- and AtTIP2;3-mediated methylammonium detoxification in yeast depended on a functional vacuole, which was in agreement with the subcellular localization of green fluorescent protein-fusion proteins on the tonoplast in planta. Transcript levels of both AtTIPs were influenced by nitrogen supply but did not follow those of the nitrogen-derepressed ammonium transporter gene AtAMT1;1. Transgenic Arabidopsis plants overexpressing AtTIP2;1 did not show altered ammonium accumulation in roots after ammonium supply, although AtTIP2;1 mRNA levels in wild-type plants were up-regulated under these conditions.   The sulfur requirement of plants varies strongly between species and can be defined as relative growth rate times the plants' sulfur content. In gen-eral sulfate taken up by the roots is the major sulfur source for growth, which has to be reduced to sul-fide prior to its metabolism into essential sulfur compounds. Plants are also able to metabolize fo-liarly absorbed sulfur gases as sulfur source for growth. The reduction of sulfur takes predominantly place in the shoot in the chloroplast. Cysteine is the precursor or sulfur donor for most other organic sulfur compounds in plants. Sulfur amino acids cys-teine and methionine are of great significance in the structure, conformation and function of proteins and enzymes. Cysteine is the precursor of glutathione, a water-soluble thiol compound which functions in the protection of plants against oxidative stress, heavy metals and xenobiotics.   The Department received 17 proposals requesting over $1,300,000 in financial assistance, for consideration during the October 2004 round of funding.   This report covers the period November 1 1999 - April 30 2000. It contains details of work carried out in field, glasshouse and laboratory trials and of staff, travel and meetings. However, due to institutional delays in the purchase of equipment, little analytical work has been done on laboratory-grown samples at either PAU Ludhiana or at NWFPAU Peshawar. A total of twenty four field trials, both on research stations and on farmers' fields, as well as seven farmer participatory trials, have been conducted, although one of the field trials again failed due to extremely high salinity. A number of laboratory and glasshouse trials have also been carried out, as well as the further crossing and selection of promising genotypes and additional germplasm collection. Administration has proceeded well, although there are still problems communicating with India due to the lack of an e-mail link.   Kim Ames (Ames 4K Farms), with his father and two brothers, operates a livestock and 3500 acre cash grain operation in Putnam County, Indiana. They have a wide variety of soils from Ragsdale-Reesville depressional poorly drained to Xenia- Russell-Miami HEL with a significant acreage of Chagrin loamy flood plain soils. They utilize as much no-till as possible but have situations which can be barriers to success. Some of these include flooding, manure injection, specialty corn, poorly drained soils and the need to grow corn after corn. They have invested a great deal of capital and management to make no-till work on as many acres as possible. Tile, lime, fertility, buffers and equipment are major components of these investments. Their no-till corn is a modified system utilizing a Phillips Harrow a few days prior to planting. Yields are higher then average for their soils.
